Peptide bioregulators: a proposed regulatory model

Very short peptides, a specific hypothesis about gene regulation, and a body of evidence that must be described accurately rather than enthusiastically.

What the term designates

In the literature, bioregulator denotes very short peptides — frequently two to four amino acids — investigated for a possible role in tissue-specific regulation of gene expression.

The proposed model is specific: a small molecule may interact with regulatory structures inside the cell and thereby modulate the activity of particular genes. Stated as a hypothesis, this is a coherent research question. Stated as a fact, it overreaches what has been shown.

Bioregulation is not the same claim

Bioregulation describes the coordination of cellular and tissue activity through the organism's own signalling systems. It is a well-established physiological concept, independent of any particular substance.

The step requiring caution is the inference: that regulatory systems exist does not establish that an externally supplied short peptide acts on them predictably, at a defined site, or with a reproducible outcome.

How they differ from receptor-binding peptides

Peptides studied for receptor interaction are usually longer, and their mechanism is described in terms of binding affinity and selectivity. Bioregulators are much shorter, and the proposed mechanism sits at the level of gene regulation rather than receptor occupancy.

Both groups share one practical constraint: enzymatic degradation and short half-life condition any interpretation of published results.

The state of the evidence

A considerable proportion of the published material on peptide bioregulators is preclinical, originates from a limited number of research groups, or appears in trade publications rather than peer-reviewed journals.
